The National Physical Laboratory (NPL) requires automation to facilitate the purification of peptides which have been synthesized via 9-Fluorenylmethoxycarbonyl (Fmoc) Solid Phase Peptide Synthesis (SPPS). Due to the intrinsic nature of this synthetic technique, it is quite common to isolate crude peptides as a mixture of the desired product and closely related impurities (e.g. protecting group adducts / deletion sequences). NPL tends to synthesize difficult peptides (self assembling, repeating motifs, slightly insoluble sequences) on 0.1 mmol scale via microwave assisted Fmoc SPPS; yielding 150 mg to 400 mg of crude product after resin cleavage. NPL is seeking to purchase a preparative HPLC, which will facilitate the purification of larger quantities of crude peptide per run (than we are currently able, i.e. >> 15-20 mg per un); and therefore yield greater quantities of highly pure peptides.
